Late third instar wing imaginal discs were cultured in Shields and Sang M3 media (Sigma) supplemented with 2% FBS (Sigma), 1% pen/strep (Gibco), 3ng/ml ecdysone (Sigma) and 2ng/ml insulin (Sigma).
